Mutue Toyota Fujii, do N\u00facleo de Pesquisa em Ficologia (IBt), que presidiu a Sess\u00e3o na qualidade de orientadora.<\/span><\/p>\n<hr align=\"center\" noshade=\"noshade\" size=\"1\" width=\"100%\" \/>\n<h3 style=\"text-align: center\">Resgate hist\u00f3rico e etiqueta molecular dos esp\u00e9cimes tipo de Ceramiales (Rhodophyta) da costa brasileira, depositados nos herb\u00e1rios SP e SPF<\/h3>\n<hr align=\"center\" noshade=\"noshade\" size=\"1\" width=\"100%\" \/>\n<h4 style=\"text-align: center\"><strong>ABSTRACT<\/strong><\/h4>\n<p><span style=\"font-weight: 400\">The gap regard to historical collections of algae and the difficulty of accessing the holotypes, coupled with limited descriptions and illustrations, result in enormous difficulties in species identification. To promote and facilitate access to these historical materials, biological collections are being cataloged in several countries and made available on online platforms. This virtual rescue is a tool to facilitate taxa comparisons and avoid the propagation of taxonomic errors. Among 107 holotype specimens of the seaweeds described from Brazil, 23 taxa belong to Ceramiales are deposited in the herbaria of Intituto de Bot\u00e2nica (SP) and the Universidade de S\u00e3o Paulo (SPF). The aim of this study was the historical review of the type specimens of Ceramiales deposited in these herbaria, providing images, as well as morphological and molecular information. The holotypes were examined and photographed in a standardized way. In order to obtain samples for new morphological and molecular studies, new collections were carried out in the type localities. The samples were collected manually, preserved in formaldehyde at 4% for morphological studies and\/or stored in desiccant silica gel for molecular studies. Among the 23 holotypes of Ceramiales, Rhodomelaceae and Ceramiaceae were the most representative families with 10 and 6 taxa, respectively. One of the great challenges inherent to this work was to rediscover taxa in the type locality to obtain DNA due to the small size of many species and the rare occurrence in the Brazilian flora, in addition to the environmental changes occurred in the last decades. This fact prevented obtaining molecular tags for 15 taxa. However, it was possible to evaluate the phylogenetic position of parasitic algae,\u00a0<\/span><i><span style=\"font-weight: 400\">Spyridiocolax capixaba<\/span><\/i><span style=\"font-weight: 400\">, an endemic species from Brazil, which is poorly known. This is the first molecular characterization of the genus\u00a0<\/span><i><span style=\"font-weight: 400\">Spyridiocolax<\/span><\/i><span style=\"font-weight: 400\">\u00a0and the data generated will contribute to the knowledge about this group of parasitic red algae. The results obtained with several historical information compiled will serve as a basis for surveys of biodiversity through the availability of these data to the phycologists and validation of the taxonomic identity of these specimens, often rare, described from Brazilian coast.<\/span><br \/>\n<strong>Keywords<\/strong>:\u00a0 <span style=\"font-weight: 400\">Ceramiales, historical collection, molecular tag, holotype, macroalgae, Rhodophyta, taxonomy.<\/span><\/p>\n<p>&nbsp;<\/p>\n<h4 style=\"text-align: center\"><strong>RESUMO<\/strong><\/h4>\n<p style=\"text-align: justify\"><span style=\"font-weight: 400\">A dificuldade de acesso e lacunas no conhecimento das cole\u00e7\u00f5es hist\u00f3ricas e hol\u00f3tipos de algas, aliadas \u00e0s limitadas descri\u00e7\u00f5es e ilustra\u00e7\u00f5es, acarretam enormes dificuldades na identifica\u00e7\u00e3o das esp\u00e9cies. P<\/span><span style=\"font-weight: 400\">ara promover e facilitar o acesso a esses materiais hist\u00f3ricos, as cole\u00e7\u00f5es biol\u00f3gicas est\u00e3o sendo catalogadas em diversos pa\u00edses e disponibilizadas em plataformas \u201conline\u201d. Esse<\/span> <span style=\"font-weight: 400\">resgate virtual \u00e9 de extrema import\u00e2ncia para facilitar a compara\u00e7\u00e3o dos t\u00e1xons e evitar a perpetua\u00e7\u00e3o de erros taxon\u00f4micos. Dentre os 107 tipos nomenclaturais de algas marinhas vermelhas descritas a partir da ficoflora brasileira, 23 t\u00e1xons pertencentes \u00e0 ordem Ceramiales est\u00e3o depositados nos herb\u00e1rios do Instituto de Bot\u00e2nica (SP) e da Universidade de S\u00e3o Paulo (SPF). Este trabalho teve como objetivo realizar o resgate hist\u00f3rico dos esp\u00e9cimes tipos de Ceramiales depositados nestes herb\u00e1rios, disponibilizando imagens, assim como informa\u00e7\u00f5es morfol\u00f3gicas e moleculares. Os hol\u00f3tipos depositados nos referidos herb\u00e1rios foram examinados, fotografados e organizados de maneira padronizada. Com o intuito de obter amostras para novos estudos morfol\u00f3gicos e moleculares, sempre que poss\u00edvel e necess\u00e1rio, as localidades tipo foram visitadas. Nesses casos, as algas foram coletadas manualmente, preservadas em formol a 4% para estudos morfol\u00f3gicos e armazenadas em s\u00edlica gel dessecante para estudos moleculares. Dentre os 23 hol\u00f3tipos de Ceramiales, Rhodomelaceae e Ceramiaceae foram as fam\u00edlias mais representativas, com 10 e seis t\u00e1xons, respectivamente. Um dos grandes desafios inerentes a este trabalho foi reencontrar t\u00e1xons nas localidades tipo para a obten\u00e7\u00e3o de DNA devido ao pequeno porte de muitas esp\u00e9cies e ocorr\u00eancia rara na flora brasileira, al\u00e9m das altera\u00e7\u00f5es ambientais ocorridas nas \u00faltimas d\u00e9cadas. Tal fato impediu a obten\u00e7\u00e3o de etiquetas moleculares para 15 t\u00e1xons. Contudo, foi poss\u00edvel avaliar a posi\u00e7\u00e3o filogen\u00e9tica da alga parasita, <\/span><i><span style=\"font-weight: 400\">Spyridiocolax capixaba<\/span><\/i><span style=\"font-weight: 400\">, uma esp\u00e9cie end\u00eamica do Brasil, que \u00e9 pobremente conhecida. Foram geradas sequ\u00eancias do gene <\/span><i><span style=\"font-weight: 400\">rbc<\/span><\/i><span style=\"font-weight: 400\">L tanto para a esp\u00e9cie parasita quanto para a hospedeira, <\/span><i><span style=\"font-weight: 400\">Spyridia clavata<\/span><\/i><span style=\"font-weight: 400\">. Esta \u00e9 a primeira caracteriza\u00e7\u00e3o molecular do g\u00eanero <\/span><i><span style=\"font-weight: 400\">Spyridiocolax<\/span><\/i><span style=\"font-weight: 400\"> e os dados gerados contribuir\u00e3o para melhor conhecimento acerca deste grupo de algas.
