V\u00edvian Tamaki, do N\u00facleo de Pesquisa em Plantas Ornamentais (IBt) e contou com a participa\u00e7\u00e3o da Dra. Maria Aurineide Rodrigues, do Instituto de Bioci\u00eancias da Universidade de S\u00e3o Paulo (USP), Dra. Elisa Mitsuko Aoyama, da Universidade Federal do Espirito Santo, do Dr. Rog\u00e9rio Mamoru Suzuki do N\u00facleo de Pesquisa Orquid\u00e1rio de S\u00e3o Paulo (IBt) e da Dra Silvia Ribeiro de Souza, do N\u00facleo de Ecologia (IBt).<\/p>\n<hr align=\"center\" noshade=\"noshade\" size=\"1\" width=\"100%\" \/>\n<h3 style=\"text-align: center\">Assimila\u00e7\u00e3o de diferentes fontes nitrogenadas na indu\u00e7\u00e3o da multiplica\u00e7\u00e3o da brom\u00e9lia <em>Alcantarea imperialis<\/em> (Carri\u00e8re) Harms cultivada <em>in vitro<\/em><\/h3>\n<hr align=\"center\" noshade=\"noshade\" size=\"1\" width=\"100%\" \/>\n<h4 style=\"text-align: center\"><strong>ABSTRACT<\/strong><\/h4>\n<p style=\"text-align: justify\">Most Bromeliaceae is considered ornamental, becoming illegal extraction target, for example, <em>Alcantarea imperialis<\/em> (Carriere) Harms. This species is endangered, with the aggravating its slow growth to reach reproductive age and their inability to propagate vegetatively under natural conditions. The <em>in vitro<\/em> culture, along with nutritional studies, has been used in the conservation of many species of economic interest, since it enables the propagation and optimization of its growth. One of the key nutrients for plant growth is nitrogen which is available mainly in the form of nitrate and ammonium, which can be provided separately or in combination to the culture medium. Preliminary studies at the Research Center for Ornamental Plants of IBt, have shown that changes in Murashige and Skoog (MS), as the reduction in the concentration of N favors the multiplication of <em>A<\/em>.<em> imperialis<\/em>. However, studies on the isolated supply of different nitrogen sources and vegetative propagation of this species are non-existent. Thus the objective of this project was to determine the best nitrogen source for multiplication and <em>in vitro<\/em> growth of <em>A<\/em>.<em> imperialis<\/em>, and through anatomical studies, to understand the development of buds responsible for the formation of shoots in order to provide grants for studies propagation and consequently to conserve species. Thus, in this study, in vitro germinated seedlings were transferred to MS medium (four concentrations and three nitrogen sources). Three samples were taken, one at 30 days after seed deposition in the culture medium and 3 and 6 months after the transfer of seedlings for treatments. The plants were analyzed for nitrogen assimilation and for their growth. After defining the ideal culture medium for multiplication and growth of plants, anatomical studies for the study of buds and roots were conducted. According to the results, the nitrate is considered the best source of nitrogen when compared to ammonium metabolized at all sampling times. Nitrate is related to the multiplication of plants, as well as mitigate the effects of ammonium toxicity, may be signaling the production of cytokines, a related hormone with the formation of shoots. Regarding the anatomical analysis it was not possible keep up with the ontogenesis of buds in both culture media of 15 mM NO<sub>3<\/sub><sup>&#8211;<\/sup> (control) and in the 30 mM NH<sub>4<\/sub>NO<sub>3<\/sub> (multiplication inductor), but the control media was the most favorable for maintenance of structural organization of the roots. It follows therefore that the species under study is multiplied when there is a combination of nitrate and ammonium, particularly at a concentration of 30 mM NH<sub>4<\/sub>NO<sub>3<\/sub>.<br \/>\n<strong>Keywords<\/strong>: Bromeliaceae, conservation, nitrogen<\/p>\n<h4 style=\"text-align: center\"><strong>RESUMO<\/strong><\/h4>\n<p style=\"text-align: justify\">A maioria dos representantes de Bromeliaceae \u00e9 considerada ornamental, tornando-se alvo do extrativismo ilegal como, por exemplo, <em>Alcantarea imperialis<\/em> (Carri\u00e8re) Harms. Essa esp\u00e9cie encontra-se amea\u00e7ada de extin\u00e7\u00e3o, tendo como agravante o seu lento crescimento para atingir a idade reprodutiva e a sua incapacidade de se propagar vegetativamente em condi\u00e7\u00f5es naturais. O cultivo <em>in vitro,<\/em> juntamente com os estudos nutricionais, tem sido utilizado na conserva\u00e7\u00e3o de diversas esp\u00e9cies de interesse econ\u00f4mico, visto que possibilita a propaga\u00e7\u00e3o e a otimiza\u00e7\u00e3o do seu crescimento. Um dos principais nutrientes para o crescimento das plantas \u00e9 o nitrog\u00eanio que se encontra dispon\u00edvel, principalmente, nas formas de nitrato e de am\u00f4nio, os quais podem ser fornecidos separadamente ou combinados ao meio de cultura. Dados preliminares, obtidos no N\u00facleo de Pesquisa em Plantas Ornamentais do IBt, mostraram que modifica\u00e7\u00f5es no meio de Murashige &amp; Skoog (MS), como a redu\u00e7\u00e3o na concentra\u00e7\u00e3o de N, favorece a multiplica\u00e7\u00e3o de <em>A<\/em>. <em>imperialis<\/em>. No entanto, estudos sobre o fornecimento isolado de diferentes fontes nitrogenadas e da multiplica\u00e7\u00e3o vegetativa dessa esp\u00e9cie s\u00e3o inexistentes. Com isso o objetivo deste projeto foi determinar a melhor fonte nitrogenada para a multiplica\u00e7\u00e3o e o crescimento <em>in vitro<\/em> de <em>A<\/em>. <em>imperialis<\/em>, e por meio de estudos anat\u00f4micos, compreender o desenvolvimento das gemas respons\u00e1veis pela forma\u00e7\u00e3o dos brotos a fim de fornecer subs\u00eddios para estudos de propaga\u00e7\u00e3o e, consequentemente, para conserva\u00e7\u00e3o da esp\u00e9cie. Desse modo, no presente estudo, pl\u00e2ntulas germinadas <em>in vitro<\/em> foram transferidas para meio MS modificado (quatro concentra\u00e7\u00f5es e tr\u00eas fontes nitrogenadas). Foram realizadas tr\u00eas coletas, uma aos 30 dias ap\u00f3s a deposi\u00e7\u00e3o das sementes no meio de cultura e 3 e 6 meses ap\u00f3s a transfer\u00eancia das pl\u00e2ntulas para os tratamentos. As plantas foram analisadas quanto \u00e0 assimila\u00e7\u00e3o do nitrog\u00eanio e quanto ao seu crescimento. Ap\u00f3s a defini\u00e7\u00e3o do melhor meio de cultivo para a multiplica\u00e7\u00e3o e crescimento das plantas, foram realizados estudos anat\u00f4micos para o estudo das gemas e das ra\u00edzes. Segundo os resultados, o nitrato foi considerado a fonte de nitrog\u00eanio melhor metabolizada quando comparada ao am\u00f4nio, em todos os tempos de coleta. O nitrato est\u00e1 relacionado com a multiplica\u00e7\u00e3o das plantas, pois al\u00e9m de amenizar os efeitos de toxicidade do am\u00f4nio, pode estar sinalizando a produ\u00e7\u00e3o de citocininas, um horm\u00f4nio relacionado com a forma\u00e7\u00e3o de brotos. Em rela\u00e7\u00e3o \u00e0s an\u00e1lises anat\u00f4micas, n\u00e3o foi poss\u00edvel acompanhar a ontog\u00eanese das gemas em plantas cultivadas em meio de 30 mM de NH<sub>4<\/sub>NO<sub>3<\/sub> (indutor de multiplica\u00e7\u00e3o) devido \u00e0 baixa porcentagem de brota\u00e7\u00e3o, mas o meio controle foi o mais favor\u00e1vel para a manuten\u00e7\u00e3o da organiza\u00e7\u00e3o estrutural das ra\u00edzes.
