No entanto, apenas 5 a 10 % da real diversidade nestas regi\u00f5es s\u00e3o conhecidos e, portanto, numerosos morfotipos e gen\u00f3tipos devem ainda ser descobertos. Al\u00e9m da ampla diversidade destes organismos, h\u00e1 d\u00e9cadas as cianobact\u00e9rias s\u00e3o reconhecidas tamb\u00e9m por produzir grande variedade de compostos biologicamente ativos com potencial uso biotecnol\u00f3gico, no entanto, poucos estudos t\u00eam sido desenvolvidos, sobretudo em regi\u00f5es tropicais e subtropicais. Deste modo, o presente estudo teve por objetivo a caracteriza\u00e7\u00e3o da biodiversidade do complexo <em>Phormidium<\/em> em distintos habitats aqu\u00e1ticos e terrestres dos biomas Mata Atl\u00e2ntica e Pantanal.\u00a0 Al\u00e9m disso, visou a prospec\u00e7\u00e3o de compostos biologicamente ativos em linhagens selecionadas. Assim, an\u00e1lises morfol\u00f3gicas foram conduzidas de modo definir as caracter\u00edsticas diagn\u00f3sticas que permitissem a detalhada caracteriza\u00e7\u00e3o dos t\u00e1xons. A an\u00e1lise a partir do gene 16S RNAr permitiu definir dez clados filogen\u00e9ticos distintos, corroborando a heterogeneidade do complexo <em>Phormidium.<\/em> Dentre estes grupos, apenas tr\u00eas linhagens foram caracterizadas como <em>Phormidium sensu stricto<\/em>, juntamente com <em>Phormidium <\/em>cf. <em>irrigum<\/em> CCALA 759, que \u00e9 um representante do grupo taxon\u00f4mico ao qual a esp\u00e9cie-tipo do g\u00eanero (<em>Phormidium lucidum<\/em>) est\u00e1 inserida.\u00a0 Dez linhagens analisadas foram identificadas como pertencentes a g\u00eaneros j\u00e1 descritos em literatura: <em>Microcoleus<\/em>, <em>Wilmottia<\/em> e <em>Desertifilum<\/em>. Entretanto, 43% do material analisado foram considerados como novos t\u00e1xons para a ci\u00eancia, e cerca de seis novos g\u00eaneros e treze esp\u00e9cies poder\u00e3o ser descritos futuramente. Neste cen\u00e1rio, o novo g\u00eanero <em>Trichocapitatum<\/em> foi descrito neste estudo, sendo distinguido as esp\u00e9cies <em>T. Komarekianum<\/em> e <em>T. lacustre<\/em>. Al\u00e9m disso, as linhagens isoladas a partir de amostras coletadas no Pantanal e na Mata Atl\u00e2ntica apresentaram-se morfologicamente distintas, o que foi corroborado a partir da an\u00e1lise filogen\u00e9tica do gene 16S RNAr. Assim, foi poss\u00edvel constatar neste estudo que a especificidade de habitats de ambas as regi\u00f5es de estudo refletiu diretamente na diversidade biol\u00f3gica. As rela\u00e7\u00f5es interespec\u00edficas foram definidas a partir dos marcadores <em>rbcL<\/em>X e <em>rpo<\/em>C1, al\u00e9m da an\u00e1lise da estrutura secund\u00e1ria do espa\u00e7ador interg\u00eanico 16S-23S RNAr. Entretanto, o n\u00famero reduzido de sequ\u00eancias referentes a estes marcadores nos bancos p\u00fablicos impossibilitou muitas vezes a compara\u00e7\u00e3o entre t\u00e1xons filogeneticamente relacionados. Neste aspecto, o presente estudo contribuir\u00e1 significativamente com o avan\u00e7o da ci\u00eancia, uma vez que foram geradas cerca de 90 sequ\u00eancias do complexo <em>Phormidium<\/em> referentes a estes marcadores. A partir dos genes <em>nif<\/em>H e <em>nif<\/em>D, foi poss\u00edvel caracterizar 16 linhagens potencialmente diazotr\u00f3ficas, bem como corroborar os grupos filogen\u00e9ticos definidos a partir do gene 16S RNAr. Com base nos testes bioautogr\u00e1ficos verificou-se a presen\u00e7a de subst\u00e2ncias com atividade anticolinester\u00e1sica, f\u00fangica e antioxidante, bem como da cianotoxina BMAA. Em suma, o presente trabalho demonstrou a ampla diversidade de prov\u00e1veis esp\u00e9cies novas para a ci\u00eancia nos ambientes tropicais e subtropicais, o papel ecol\u00f3gico destes organismos como potenciais fixadores de nitrog\u00eanio, bem como o uso de diferentes marcadores moleculares para auxiliar na defini\u00e7\u00e3o de esp\u00e9cies no complexo <em>Phormidium.<\/em><\/p>\n<p style=\"text-align: justify\"><strong>Palavras-chaves:<\/strong> <em>Phormidium sensu lato<\/em>, taxonomia polif\u00e1sica, m\u00faltiplos-loci g\u00eanicos, compostos bioativos, linhagens diazotr\u00f3ficas.<\/p>\n<p style=\"text-align: justify\" align=\"left\"><b>Abstract<\/b><\/p>\n<p style=\"text-align: justify\">In tropical and subtropical regions, the richness of different habitats and consequent ecological diversity allow for high morphological variety of cyanobacteria. However, only 5 to 10% of the real diversity of these regions is known and, therefore, numerous morphotypes and genotypes are yet to be discovered. In addition to the ample diversity of these organisms, cyanobacteria have also been recognized, for decades, as producers of a wide variety of biologically active compounds, potentially suitable for biotechnological applications. However, few studies, especially in tropical and subtropical regions, have been carried out. Accordingly, the present study aimed to characterize the population biodiversity of the <em>Phormidium <\/em>complex in distinct aquatic and terrestrial habitats of the Atlantic rainforest and Pantanal biomes. In addition, it also prospected biologically active compounds in selected lineages. Thus, morphological analyses were carried out to define diagnostic features for detailed characterization of the taxa. The analysis based on gene 16S RNAr allowed the definition of ten distinct phylogenetic clades, corroborating the <em>Phormidium<\/em> complex heterogeneity<i>.<\/i> In these groups, only three lineages were characterized as <em>Phormidium sensu stricto<\/em>, together with <em>Phormidium <\/em>cf. <em>irrigum<\/em> CCALA 759, which is a representative of the taxonomic group in which the species-type of the genus (<em>Phormidium lucidum<\/em>) is inserted. Ten analyzed lineages belong to taxa already described in the literature, such as <em>Microcoleus<\/em>, <em>Wilmottia<\/em> and <em>Desertifilum<\/em>. However, the other analyzed lineages (43%) could be considered as new entities for science, and about six new genera and thirteen species can be described eventually.\u00a0 In this scenario, the new genus <em>Trichocapitatum<\/em> was described in this study and three distinct species, <em>T. komarekianum<\/em> and <em>T. lacustre<\/em><i>, <\/i>were characterized. Furthermore, the strains isolated from Pantanal and Mata Atlantica was morphologically distinct, which was supported by the 16S rRNA phylogenetic analysis. Thus, in this study we determined that habitats specificity from both study areas reflected directly the biological diversity. Interspecific relations were defined based on <em>rbcL<\/em>X and <em>rpo<\/em>C1 markers and the secondary structure analysis of the 16S-23S RNAr intergenic spacer. However, the small number of sequences related to these markers in public database often impossible to compare phylogenetically related taxa. In this context, this study will contribute significantly to the advancement of science, since about 90 sequences of <em>Phormidium <\/em>complex regarding these markers were generated. Based on genes <em>nif<\/em>H and <em>nif<\/em>D, it was possible to characterize 16 potentially diazotrophic lineages as well as corroborating the phylogenetic groups defined based on gene 16S RNAr.
